Ex- Employee created a file under private ownership in my Entrprise Account and is accessing it from an external source. I need to get owbership of this file and remove that user's account.

To confirm, is the content owned by an external user or a managed user of your organization? Please check out this article for the difference between these type of users.
If this content is owned by a managed user of your Box, and you have Admin privileges, as an admin you may perform 'Delete and Transfer' which would allow you to delete the managed user and transfer ownership of the contents owned by that user to you.
However, if this is a content owned of an external user, you would need to reach out to the owner and discuss the possibility of them transferring the ownership of this content to you. They would have to perform 'Ownership transfer' that is discussed in this article for you to get control of the content.
