If I copy a file to a new folder in Box, will changes in one Box location show up in the other location and vice versa?

To answer your question, no, the changes will only save to one location.
Rather than copying the file, I would suggest to use "Bookmarks" to create a quick link of that file which you can use on multiple locations in your Box.
